Preparation and Evaluation of Pirfenidone Dry Powder by Spray Drying Technology

Inhalation of pirfenidone is one the possible methods for effective treatment idiopathic pulmonary fibrosis. In this study, we attempted to prepare inhaled dry powder through reasonable design precursor formulation and parameter adjustment spray drying process. The mechanism polyvinyl alcohol (PVA) polymer as surfactant used assist dissolution in water, instead using organic solvents (such ethanol methanol) dissolution, so reduce production explosion-proof requirements avoid safety problems that may be caused by solvent residues. Leucine first enriched crystallized on surface particles then condenses into a hydrophobic shell which collapses form wrinkles cohesion between particles. When pirfenidone: alcohol: leucine=2:1:3, inlet temperature 120 °C, content active pharmaceutical ingredient (API) 40.7%, relative reaches 81.4% with fraction fine (FPF) 26.2%, represents good drug loading inhalation performance.
